BN QuickPickTM genomic Plant DNA
For the rapid purification of genomic plant DNA

    

The QuickPick genomic Plant  DNA purification kit provides a rapid and simple means of isolating genomic DNA from plant tissue.  The method does not require any organic solvents and eliminates the need for centrifugation, vacuum filtration or column separation.  

Principle
DNA in the sample is released using Plant DNA Proteinase K Solution and Plant DNA Lysis Buffer.  Released DNA is bound to Plant DNA MagaZorbTM Magnetic Particles in the presence of Plant DNA Binding Buffer.  Plant DNA MagaZorbTM Magnetic Particles with the bound DNA are captured with the PickPenTM and the contaminants are removed by washing with Plant DNA Wash Buffer.  The DNA is then eluted from the magnetic particles with the Plant DNA Elution Buffer.  The procedure starting from homogenised plant sample and ending with purified DNA lasts less than 40 minutes.

BN QuickPickTM genomic Plant DNA isolation protocol

  1. Number tubes 1 to 5 and pipette QuickPick genomic Plant DNA kit reagents into tubes as follows :
Tube 1 : 5 mL Plant DNA Proteinase K Soultion is added to the homogenised leaf tissue suspended in30 mL of the Plant DNA Lysis Buffer.  Mix by gentle pulse vortexing and lyse at 650 for 15-30 minutes.

Lyse at 650 for 15-30 minutes 
Tube 2 : 125 mL Plant DNA Binding  Buffer, 5 mL Plant DNA Magnetic Particles.
Tube 3 : 200 mL Plant DNA Wash Buffer.
Tube 4 : 200 mL Plant DNA Wash Buffer.
Tube 5 : 100 mL Plant DNA Elution Buffer

  1. Remove Tube 1 from 650 C.  Centrifuge tube at 16,000 x g for 5 minutes.  Gently transfer supernatant into Tube 2 (Plant DNA Binding Buffer. Plant DNA MagaZorbTM Magnetic Particles).  Mix tube gently and incubate at room temperature for 2-10 minutes. 

Bind at room temperature with continuous mixing for 2-10 minutes
  1. Collect the magnetic particles from Tube 2 with the PickPen and release them into Tube 3 (Plant DNA Wash Buffer).  Repeat washing step in Tube 4.
  1. Collect the magnetic particles from Tube 4 with the PickPen and release them into Tube 5 (Plant DNA Elution Buffer).  Mix Tube 5 gently and continuously and elute at room temperature for 5 - 10 minutes.   

  1. Collect the magnetic particles from Tube 5 and discard them.  The eluate containing genomic DNA is ready for further analysis.

        3.    Collect the magnetic particles from Tube 2 with the PickPen and release them into Tube 3 (Plant DNA Wash Buffer).  Repeat
             washing step in Tube 4.

       4.     Collect the magnetic particles from Tube 4 with the PickPen and release them into Tube 5 (Plant DNA Elution Buffer). 
                Mix Tube 5 gently and continuously and elute at room temperature for 5 - 10 minutes.        

        

Elute at room temperature with continuous mixing for 5 - 10 minutes

        5.     Collect the magnetic particles from Tube 5 and discard them.  The eluate containing genomic DNA is ready for further analysis.
