BN QuickPickTM gDNA
For the rapid purification of genomic DNA

    

The QuickPick gDNA purification kit provides a rapid and simple means of isolating genomic DNA from whole blood, buffy coat, leucocytes and cultured cells. The kit is suitable for use with whole blood which has been anticoagulated with commonly used anticoagulants such as EDTA, heparin and citrate.  The method does not require any organic solvents and eliminates the need for centrifugation, vacuum filtration or column separation.  DNA purified using the QuickPick gDNA purification kit is of high molecular weight and it is suitable for downstream applications such as different PCR amplifications, restriction enzyme digestions and sequencing.

Principle
DNA in the sample is released using gDNA Proteinase K solution and gDNA Lysis Buffer.  Released DNA is bound to gDNA MagaZorbTM Magnetic Particles in the presence of gDNA Binding Buffer.  gDNA MagaZorbTM with the bound DNA are captured by PickPen and contaminants are removed by washing with gDNA Wash Buffer.  The DNA is eluted from the magnetic particles with the gDNA Elution Buffer.  The procedure starting from whole blood to pure DNA takes less than 20 minutes.

Number tubes 1 to 5 
Tube 1
: 50
mL gDNA Lysis Buffer, 50 mL sample, 5 mL Proteinase K Solution.
Tube 2 : 125
mL gDNA Binding Buffer, 5 mL gDNA Magnetic Particles.
Tube 3 : 200
mL gDNA Wash Buffer.
Tube 4 : 200
mL gDNA Wash Buffer. 
Tube 5
: 200
mL gDNA Elution Buffer

Protocol
  1. Mix Tube 1 by pulse-vortexing for 15 seconds.
  1. Incubate Tube 1 (gDNA Lysis Buffer, sample, gDNA Proteinase K Solution) at 560 for at least 10 minutes.
Incubate at 560 for at least 10 minutes
  1. Remove Tube 1 from the heating block. Pipette the whole suspension from Tube 1 into Tube 2 (gDNA Binding Buffer, gDNA MagaZorb Magnetic Particles)        

Incubate at room temperature for at least 2 minutes

  1. Mix Tube 2 gently and incubate at room temperature for at lease 2 minutes. During this step the DNA binds to the magnetic particles
  1. Collect the magnetic particles from Tube 2 with the PickPen and release them into  Tube 3 (gDNA Wash Buffer)
  1. Collect the magnetic particles from Tube 3 with PickPen and release them into Tube 4  (gDNA Wash Buffer).
  1. Collect the magnetic particles from Tube 4 with PickPen and release them into Tube 5 (gDNA Elution Buffer) Mix gently and incubate at room temperature for at least 2 minutes.  Genomic DNA is now eluted into the solution.
Incubate at room temperature for at least 2 minutes
  1. Collect the magnetic particles from Tube 5 and discard.  The eluate containing the DNA is ready for further analysis.
